When I did my 10 gallon, I had the glass professionally cut (I could never get it to break right when I tried). Then I basically just took some fine sand paper and scuffed up the areas where the joints would be (so the silicone would stick). Then I layed out the bottom, put a bead of silicone down and attached the back pane using a couple strips of electrical tape to hold it. Then I did the sides, then the front in the same manner. I added another bead of silicone on all the inside seams and smoothed it out with my finger.
I let it dry for 24 hours and filled it with water to see if it would leak.

Another set of hands would have been helpful to hold the panes in place while I applied the electrical tape, but other than that it wasn't too difficult.

Here's a good site that will even give you the dimensions to have the glass cut. There is also some interesting articles on building plywood tanks with glass panels (for built-in aquariums). http://www.garf.org/
